manufacturing, and healthcare isolation facilities, differential pressure control is a mandatory
cornerstone. So, what is the required cleanroom differential pressure level? How do positive and
negative pressure principles function? Where should gauges and sensors be positioned to prevent
reading errors?
1. Why Must Cleanrooms Maintain Differential Pressure Control?
Differential pressure is the difference in static pressure between two adjacent areas (e.g., between a cleanroom and a corridor, or between two cleanrooms of different cleanliness classes). The primary purpose of maintaining differential pressure is to ensure airflow follows a strictly defined direction, preventing cross-contamination from airborne particles and microorganisms.

Positive Pressure Principle: Applied in rooms where products require ultimate protection from contamination (e.g., sterile pharmaceutical compounding, aseptic packaging, semiconductor fabrication). The cleaner room maintains HIGHER pressure than surrounding areas. When doors open, clean air flows outward, preventing external dust and microbes from entering.

Negative Pressure Principle: Applied in containment rooms handling hazardous substances, volatile chemicals, vaccine production, or infectious disease isolation. Pressure inside the room is kept LOWER (negative) relative to adjacent spaces. 2. What is the Standard Cleanroom Differential Pressure Range?
While there is no single static number for every application, international standards such as ISO 14644-4 and EU GMP / WHO GMP guidelines recommend the following operational targets:

Standard Benchmark Range: 10 Pa to 15 Pa (Pascals) differential between adjacent spaces of different cleanliness classifications.

Design Flexibility Range: 5 Pa to 20 Pa depending on the total number of cascading pressure steps, room dimensions, and door-opening frequencies.

Airlock Pressure Cascades: Typically, a 10–15 Pa pressure drop is maintained between the primary cleanroom and the airlock, and an additional 10–15 Pa between the airlock and the unclassified corridor. Consequently, the primary cleanroom stays 20–30 Pa higher than the outer corridor.
3. Engineering Guidelines for Monitoring Equipment Placement
To guarantee continuous pressure stability and immediate alarm response during pressure loss, monitoring devices should be deployed in a tiered architecture:
